目的 观察水杉总黄酮 (TFM)对容量超负荷大鼠心肌肥厚的作用及其对心室 c- Fos蛋白表达的影响。方法 采用腹腔动 -静脉造瘘建立大鼠容量超负荷模型 ,ig给予 TFM,免疫组化测定心室 c- Fos蛋白的相对含量。结果 TFM剂量依赖性地减轻大鼠心脏重量 ,抑制心室 RNA和蛋白质合成 ,并抑制心室 c- Fos蛋白表达。结论 TFM可预防容量超负荷大鼠心肌肥厚的形成 ,抑制心室 c- Fos蛋白表达可能是其分子机制之一。
Objective To observe the effect of total flavonoids of Metasequoia glyptostroboides (TFM) on cardiac hypertrophy in rats with volume overload and its effect on the expression of c-Fos protein in ventricles. Methods Rat model of volume overload was established by peritoneal arteriovenous fistula. ig was given to TFM. Immunohistochemistry was used to determine the relative content of c-Fos protein in ventricles. Results TFM dose-dependently reduced cardiac weight, inhibited ventricular RNA and protein synthesis, and inhibited c-Fos protein expression in the ventricles. Conclusion TFM can prevent the formation of cardiac hypertrophy in rats with volume overload. The inhibition of c-Fos protein expression in the ventricle may be one of its molecular mechanisms.
