目的探讨颌突上皮和成釉器上皮在E12.5颌突外胚间充质细胞向牙齿样结构形成中的作用。方法切取E12.5大鼠下颌突,将纯化的外胚间充质细胞(A组)、颌突上皮细胞+外胚间充质细胞(B组)、成釉器上皮+外胚间充质细胞(C组)、成釉器上皮(D组)与明胶海绵支架复合,分别植入大鼠肾被膜下,4周后收获移植物,对移植物进行组织学观察。结果种植4周,颌突上皮+外胚间充质细胞在肾被膜下体积长大,形成牙本质-牙髓复合体样结构。成釉器上皮+外胚间充质细胞在肾被膜下体积增大,形成含牙釉质、牙本质-牙髓复合体样结构。成釉器上皮形成类骨样结构,纯化的外胚间充质细胞团植入肾被膜下后不能生长,细胞团块被吸收消失。结论牙源性上皮(颌突上皮和成釉器上皮)在E12.5颌突外胚间充质细胞向牙齿样结构形成中起决定作用,晚期牙源性上皮(成釉器上皮)可能仍蕴含成牙信息。
Objective To investigate the role of maxillary epithelium and enamel epithelium in the formation of dental structures of E12.5 jugular ectomesenchymal cells. Methods The edentulous mandibles of E12.5 rats were dissected, and purified ectomesenchymal cells (A group), jugular epithelial cells + ectomesenchymal cells (B group), enamel epithelium + ectomesenchyma Cells (group C) and enamel epithelium (group D) were combined with gelatin sponge scaffolds and implanted into rat renal capsule respectively. After 4 weeks, the grafts were harvested for histological observation. Results After 4 weeks of implantation, the jugular epithelium + ectomesenchymal cells grew up under the renal capsule, forming dentin-pulp complex-like structures. Enamel epithelium + ectomesenchymal cells in the renal capsule volume increases, the formation of enamel, dentin - pulp like complex structure. The enamel-like epithelium forms a bone-like structure. The purified ectomesenchymal cell mass can not grow under the renal capsule, and the cell mass is absorbed and disappeared. Conclusions Odontogenic epithelium (jugular epithelium and enamel epithelium) play a decisive role in the formation of tooth-like structures in e12.5 jugular ectomesenchymal cells.
