目的:探讨天麻素在体外对人胃癌细胞SGC-7901细胞活力和凋亡的影响。方法:培养人胃癌细胞株SGC-7901,随机分为正常组、对照组和实验组,再按不同时间点分为6、12、24、48、72 h 5个亚组,每个亚组8孔细胞。实验组按2 mg/m L加入天麻素,对照组加入等量生理盐水,正常组不作其它处理。运用MTT法检测不同时间点细胞活力,运用TUNEL法检测细胞凋亡情况。结果:天麻素对人胃癌细胞SGC-7901具有增殖抑制和促进凋亡作用,并具有时间依赖关系。正常组、对照组之间细胞活力及凋亡比较均无统计学意义(P>0.05)。在6 h、12 h、24 h、48 h、72 h时间点,实验组的细胞活性分别为0.41±0.05、0.42±0.07、0.43±0.06、0.47±0.07、0.52±0.08,对照组分别为0.44±0.07、0.49±0.12、0.50±0.09、0.69±0.11、0.72±0.12,实验组细胞活力较对照组降低(P<0.05),实验组的凋亡指数分别为7.12±0.65、10.48±1.26、15.37±1.69、17.52±2.33、33.62±3.65,对照组的凋亡指数分别为1.08±0.05、3.41±0.56、6.09±0.91、9.52±1.14、11.57±1.18,实验组较对照组凋亡显著增加(P<0.05)。结论:天麻素可降低人胃癌SGC-7901细胞的活力并增加细胞的凋亡,可能具有抗肿瘤的作用。
Objective: To investigate the effect of gastrodin on the viability and apoptosis of human gastric cancer cell line SGC-7901 in vitro. Methods: The human gastric cancer cell line SGC-7901 was cultured and divided into normal group, control group and experimental group randomly, and divided into 5 subgroups at 6, 12, 24, 48 and 72 h at different time points. Each subgroup 8 Hole cells. In the experimental group, gastrodin was added at 2 mg / m L, normal saline was added in the control group, and no treatment was given in the normal group. Cell viability was measured by MTT assay at different time points, and apoptosis was detected by TUNEL assay. Results: Gastrodin could inhibit proliferation and promote apoptosis of human gastric cancer cell SGC-7901 in a time-dependent manner. There was no significant difference in cell viability and apoptosis between normal group and control group (P> 0.05). At 6 hours, 12 hours, 24 hours, 48 hours and 72 hours, the cell viability in the experimental group was 0.41 ± 0.05, 0.42 ± 0.07, 0.43 ± 0.06, 0.47 ± 0.07 and 0.52 ± 0.08, respectively, while the control group was 0.44 ± 0.07,0.49 ± 0.12,0.50 ± 0.09,0.69 ± 0.11,0.72 ± 0.12, the cell viability in the experimental group was lower than that in the control group (P <0.05), and the apoptosis index in the experimental group was 7.12 ± 0.65,10.48 ± 1.26,15.37 ± 1.69, 17.52 ± 2.33 and 33.62 ± 3.65, respectively. The apoptotic indexes in the control group were 1.08 ± 0.05, 3.41 ± 0.56, 6.09 ± 0.91, 9.52 ± 1.14 and 11.57 ± 1.18 respectively, and the apoptotic index in the experimental group was significantly higher than that in the control group <0.05). Conclusion: Gastrodin can reduce the vitality of human gastric cancer SGC-7901 cells and increase cell apoptosis, which may have anti-tumor effect.
