目的 在人成纤维细胞的生物学特性观察研究中 ,进行成纤维细胞的原代培养 ,必须获得足够量的成纤维细胞。方法 我们采用整形外科断层取皮技术 ,使用细胞刮刀 ,可相对简单获取有活力的成纤维细胞 ,并应用台盼蓝细胞活力染色排除法进行检测。结果 使用该方法可简单、经济获取足够量的成纤维细胞 ,使得短期内即可获得大量的传代细胞。1× 2cm2 的断层皮片可达到的细胞的数量级 10 6的左右。结论 断层皮片成纤维细胞培养方法 ,结合整形外科的技术 ,是一种新的、经济和快速原代成纤维细胞培养方法 ,较胰酶消化法和组织块贴壁法有明显的优越性 ,对于进行成纤维细胞移植 ,更值得加以推广和应用。
Purpose To observe the biological characteristics of human fibroblasts, primary culture of fibroblasts must be performed to obtain a sufficient amount of fibroblasts. Methods We used plastic surgery tomography, using a cell scraper to obtain relatively viable fibroblasts relatively easily and were assayed by trypan blue exclusion. Results Using this method, a sufficient amount of fibroblasts can be obtained simply and economically, so that a large number of passage cells can be obtained in a short period of time. 1 × 2cm2 of the cutaneous flap can reach the order of about 10 6 cells. Conclusion The method of culturing cutaneous fibroblasts combined with plastic surgery is a new, economical and rapid method of primary fibroblast culture. Compared with the method of pancreatic digestion and tissue block adhesion, it has obvious advantages. For fibroblast transplantation, more worthy of promotion and application.
