目的比较NP方案加同期照射对p53表达情况不同的两种人肺腺癌细胞凋亡率的影响,为临床治疗提供参考。方法按p53表达情况不同分A、B两组,A组采用表达p53的A549细胞为研究对象,B组采用不表达p53的H1299细胞为研究对象,进行细胞凋亡实验。通过MTT比色法,流式细胞仪等检测方法观察记录两组肿瘤细胞的凋亡率并进行比较分析。结果在8Gy照射水平,第12h点,A组凋亡率为2.20%,B组为2.51%,两组间差异较小,经卡方检验,P>0.05;在第24点,A组凋亡率为41.48%,B组为56.37%,两组间差异显著(P<0.05);在第48h点,A组凋亡率为74.66%,B组为93.48%,两组间差异显著(P<0.05)。结论 NP方案加同期照射对不表达p53的肿瘤细胞凋亡率明显高于表达p53的肿瘤细胞,且随时间延长凋亡率增加。
Objective To compare the effects of NP and concurrent irradiation on the apoptosis of two kinds of human lung adenocarcinoma cells with different expression of p53, so as to provide a reference for clinical treatment. Methods According to the expression of p53, A and B groups were divided into two groups. A group was treated with p53-expressing A549 cells. In group B, H1299 cells that did not express p53 were used as experimental group. The apoptosis rate of the two groups of tumor cells was observed by MTT colorimetric assay and flow cytometry and compared. Results At the 8th day, the apoptotic rate was 2.20% in group A and 2.51% in group B at 12h. There was no significant difference between the two groups (P> 0.05) The rate was 41.48% in group B and 56.37% in group B, with a significant difference between the two groups (P <0.05). At 48h, the apoptotic rate in group A was 74.66% and in group B 93.48% 0.05). Conclusion The apoptosis rate of tumor cells not expressing p53 is significantly higher than that of p53 expressing tumor cells treated with NP and radiation, and the apoptosis rate increases with time.
