目的 探讨沙鼠短暂前脑缺血脑内转化生长因子 βⅠ、Ⅱ型受体基因的表达。 方法 夹闭蒙古沙鼠双侧颈总动脉 10min诱导前脑缺血 ,原位杂交检测缺血再灌后不同时点脑内TβRⅠ、ⅡmRNAs的表达。结果 各组沙鼠脑组织中神经元和胶质细胞胞浆均可见TβRⅠ ,ⅡmRNAs阳性表达。缺血 10min后 1d、3d、7d时TβRⅠmRNA表达增加 ,而TβRⅡmRNA在 1d、3d时表达增加 ,7d时下降 (与对照组比 ,P <0 0 1)。结论 短暂前脑缺血诱导脑组织TβRImRNA表达增加 ;第 1、3dTβRIImRNA表达增加 ,但第 7d下降。提示沙鼠短暂前脑缺血第 1、3d脑组织TβRⅠ ,ⅡmRNAs表达增加 ,可能有利于TGFβ信息传递 ;第 7d低量的TβRⅡmRNA表达 ,则可能不利于TGFβ信息传递
Objective To investigate the expression of transforming growth factor β Ⅰ and type Ⅱ receptors in gerbil after transient forebrain ischemia. Methods The bilateral common carotid arteries of Mongolian gerbils were occluded for 10 min to induce forebrain ischemia. The expression of TβR Ⅰ and Ⅱ mRNA in the brain at different time points after ischemia / reperfusion was detected by in situ hybridization. Results The positive expression of TβRⅠand ⅡmRNAs in the neurons and glial cells in the brain tissue of each group were observed. The TβRⅠmRNA expression increased at 1d, 3d and 7d after ischemia 10min, while the expression of TβRⅡmRNA increased at 1d and 3d and decreased at 7d (P <0.01 compared with the control group). Conclusions TβRImRNA expression increased in transient forebrain ischemia-induced brain tissue. The expression of TβRIImRNA increased on the 1st and 3rd day, but decreased on the 7th day. It is suggested that the expression of TβRⅠorⅡmRNAs may be beneficial to the transmission of TGFβ mRNA in the first and third day after the onset of transient forebrain ischemia in gerbils. The low expression of TβRⅡmRNA may be detrimental to the transmission of TGFβin the seventh day
