目的建立一种操作简单、稳定可靠、经济实用的同种异体异位心脏移植动物模型。方法单人操作。动物麻醉后迅速摘取供心备用;受体鼠肝素化,解剖出右侧颈外静脉及其分支,结扎并剪断其分支。用微血管夹钳夹近心端主干,近颅侧结扎,距该线结约1mm缝一6-0Prolene线作牵引线,剪断血管主干,牵引线引导颈外静脉穿过Cuff管,用两把显微镊将血管壁外翻在Cuff管外,5-0丝线环扎固定。类似方法处理右侧颈总动脉。供心于无名动脉跟部前壁作一小切口,将颈总动脉Cuff管插入其内,环扎固定;同样方法将颈外静脉Cuff管插入肺动脉内并环扎固定。先开放颈外静脉,再分次开放颈总动脉。结果全部移植成功,无吻合口漏血或回流受阻,全组无手术死亡,移植物复跳率100%。总手术时间45~60min,其中吻合时间仅3~8min。结论该模型是一种实用、理想、易于复制的器官移植研究动物模型。
Objective To establish a simple, stable, reliable and economical animal model of allogenic heart transplantation. Method single operation. Animals were quickly removed after anesthesia for heart; the recipient mice were heparinized, the right external jugular vein and its branches were dissected, and the branches were ligated and cut. With micro-vascular clamp around the proximal end of the main trunk, proximal cranial ligation, from the line junction about 1mm seam 6-0Prolene line as a traction line, cut off the blood vessel trunk, traction line guide external jugular vein through Cuff tube, with two significant Micro tweezers eversion of the vessel wall Cuff tube, 5-0 silk cerclage fixed. A similar approach was performed on the right common carotid artery. Provide a small incision for the anterior part of the innominate artery and the anterior part of the carotid artery, insert the Cuff tube of the common carotid artery into it, and fasten it. First open the external jugular vein, and then open the carotid artery. Results All transplants were successful. No anastomotic leakage or obstruction was found. All the patients died without surgery and the rate of graft rebound was 100%. The total operation time 45 ~ 60min, anastomotic time is only 3 ~ 8min. Conclusion The model is a practical, ideal and easy-to-reproduce organ transplantation animal model.
