利用SRAP标记对13份浙南柚类地方资源和琯溪蜜柚及芽变进行遗传多样性分析和鉴定。结果表明:平均每个引物组合可扩增出15.7条谱带,14对SRAP引物共产生220条谱带,其中多态性谱带为122条,多态率为55.4%,表明15份材料间检测到的SRAP位点多态性不高,不同引物组合可将11个基因型完全分开。聚类分析结果显示,15份柚类种质在遗传相似系数0.97处可以分为8大类,第1类群为四季柚7个优异株系,第2类群包括琯溪蜜柚及其芽变,而平阳文旦、早香柚、处红柚、红心1号土柚、红心2号土柚、酸柚分别单独为第3、4、5、6、7、8类群。四季柚选系中务城1号、务城3号、马站红心四季柚的指纹图谱有可区分的差异,说明DNA水平发生轻微变异。
SRAP markers were used to analyze and identify the genetic diversity of 13 southern Zhejiang pomelo resources and Guanxi pomelo and buds. The results showed that 15.7 bands could be amplified by each primer combination on average, and 14 pairs of SRAP primers produced a total of 220 bands, of which 122 were polymorphic bands with a polymorphism rate of 55.4%, indicating that 15 materials The detected polymorphism of SRAP loci was not high, and 11 primer combinations could completely separate 11 genotypes. Cluster analysis showed that 15 pomelo germplasms could be divided into 8 groups at the genetic similarity coefficient of 0.97, the first group was 7 elite strains of Siji grapefruit, the second group included Guanxi pomelo and its buds, The Pingyang Pomelin, as early as fragrant pomelo, Department of red pomelo, Hong heart No. 1 soil pomelo, Hongxin No. 2 soil pomelo, grapefruit pomelo groups 3, 4, 5, 6, 7, 8 alone. Four seasons pomelo selected Department of City on the 1st, City No.3, the horse heart red four seasons grapefruit fingerprints are distinguishable, indicating a slight variation in DNA levels.
