microRNA是一类长约24个碱基的非编码RNA,其广泛表达在各种生物体内,发挥重要的调节功能,在免疫系统中,亦有报道认为其参与了免疫细胞的发育、活化等。通过实时定量PCR研究发现:mir-128-2高表达在双阳性、双阴性T细胞和共同淋巴细胞祖细胞中,而在成熟T细胞和成熟B细胞中表达显著降低。为进一步探讨m i r-128-2在淋巴细胞发育中的作用,我们通过制备过表达m i r-128-2逆转录病毒,并感染小鼠骨髓干细胞制备骨髓嵌合小鼠模型。通过FACS、实时定量PCR及Northern-blot鉴定模型成功建立。进一步使用FACS分析发现mir-128-2过表达后阻碍了B细胞发育,对T细胞发育无显著影响。进一步研究发现mir-128-2过表达后并不影响B细胞的凋亡和自我增殖,而可能是阻止了CLP向PreProB细胞的发育。通过嵌合小鼠模型建立和表型分析可见mir-128-2参与了B细胞发育的调控,表现为阻碍B细胞的发育。“,”MiRNAs(miRNAs)are about 24nt non-coding RNAs which are expressed in al organisms and play important roles. In the immune system, they are involved in the development, activation of immunocytes. At present study, we found that mir-128-2 was highly expressed in DN, DP T cells and common lymphoid progenitor cells but lowly expressed in the mature T cells and B cells. By infected hemotopoitic stem cells with fresh prepared retrovirus encoding mir-128-2, we successful y established mir-128-2 bone marrow chimeric mice identified by FACS, Northern-blot and real time PCR. Further study displayed that over-expressed mir-128-2 in mice leaded to the block the B celldevelopment but not T cell. Mir-128-2 did not affect the B cellapoptosis and self-proliferation. So mir-128-2 may inhibit the common lymphoid progenitor cells development into PreproB cells.
