目的 :研究全反式维甲酸 (ATRA)对舌癌细胞株的体内诱导分化作用。方法 :人舌癌细胞株分别经下列方法处理后接种于裸鼠皮下 ,(1)未经药物处理 ,(2 )未经药物处理的舌癌细胞接种后立即行ATRA灌胃 ,(3)经 10 μmol/LATRA处理 3天或 6天的舌癌细胞 ;观察肿瘤的生长状态、倍增时间和抑瘤率 ,3周后处死动物取出瘤块行病理学检查。结果 :经A TRA处理后 ,肿瘤生长速率减慢 ,倍增时间延长和抑瘤率增高 ,光镜下治疗组和对照组未见明显变化 ,透射电镜下可见肿瘤细胞产生一定的分化 ,出现较多的凋亡细胞。结论 :以上结果表明ATRA对体内的舌癌细胞有一定的诱导分化作用。
AIM: To investigate the effect of all-trans retinoic acid (ATRA) on inducing and differentiating tongue cancer cell lines in vivo. Methods: Human tongue cancer cell lines were inoculated subcutaneously in nude mice by the following methods: (1) untreated, (2) untreated tongue cancer cells were treated with ATRA immediately after inoculation, (3) The tongue cancer cells were treated with 10 μmol / L ATA for 3 days or 6 days. The growth status, doubling time and tumor inhibition rate were observed. After 3 weeks, the animals were sacrificed for pathological examination. Results: After A TRA treatment, the tumor growth rate was slowed down, the doubling time was prolonged and the tumor inhibition rate was increased. There was no obvious change in the treatment group and the control group under the light microscope. Transmission electron microscopy showed that the tumor cells had some differentiation Of apoptotic cells. Conclusion: The above results indicate that ATRA can induce the differentiation of tongue cancer cells in vivo.
