目的:研究苯并卟啉衍生物单环酸A(BPD)光动力杀伤人鼻咽低分化鳞癌CNE2细胞株的作用机理。方法:在体外培养的CNE2细胞株中加入不同浓度的BPD后,用690nm单色光照射仪照射,照光能量分别为1.2J/cm2、2.4J/cm2、4.8J/cm23组。MTT法检测细胞活性。并收集细胞进行电镜检查。结果:在同一光照能量下,不同浓度BPD对CNE2细胞的杀伤作用随BPD剂量的增大而增强,照光能量分别为1.2J/cm2、2.4J/cm2、4.8J/cm23组,其IC50值分别为1958.6ng/ml、506.38ng/ml和343.0ng/ml。电镜检查:BPD晚光组光照15min,细胞体积已比单纯光照射组缩小,整个细胞密度增加,胞质内所有细胞器呈轻度收缩状态;2h,一些细胞胞质出现明显自溶;8h后能见到2种形式的细胞死亡:坏死和细胞凋亡;24h,主要以坏死细胞为主。结论:在同一光照能量下,对CNE2细胞的杀伤作用随BPD剂量的增大而增强,光动力作用引起的肿瘤细胞死亡存在两种时相及方式,在同一条件下出现两种不同的时相及死亡方式,可能与细胞处在不同生长阶段,代谢程度不一,吸收BPD的含量不同有关。
