小鼠巨细胞病毒PCR诊断方法的建立及其应用

目的建立小鼠巨细胞病毒(MCMV)PCR检测方法,并进行初步应用。方法根据NCBI公布的巨细胞病毒设计引物,进行PCR体系优化、敏感性、特异性测试;运用优化后的方法,对87份小鼠血清进行检测,对阳性样本进行测序分析。结果建立的小鼠巨细胞病毒PCR方法灵敏度高,特异性好,初步检测4个屏障设施的87份小鼠血清,13份为阳性,通过测序证实为小鼠巨细胞病毒,验证了此方法的可行性。结论为小鼠巨细胞病毒的快速检测提供了方法。 Objective To establish a method for the detection of mouse cytomegalovirus (MCMV) by PCR. Methods Based on the cytomegalovirus primers designed by NCBI, the optimal, sensitive and specific PCR methods were tested. Serum samples from 87 mice were tested by the optimized method, and the positive samples were sequenced. Results The mouse cytomegalovirus PCR method established was highly sensitive and specific. 87 serum samples from 4 barrier facilities were initially detected and 13 were positive. The results of sequencing confirmed the cytotoxicity against mouse cytomegalovirus. feasibility. Conclusion The method provided for the rapid detection of mouse cytomegalovirus.
