目的:研究狗脊的化学成分,探讨其对二苯代苦味基肼自由基(DPPH)的清除作用。方法:采用硅胶色谱、凝胶色谱、高效液相色谱等技术对狗脊70%乙醇提取物进行分离纯化,通过波谱方法鉴定其结构,并对化合物进行清除DPPH活性试验。结果:从狗脊70%乙醇提取物中分离得到10个成分,分别为1-O-咖啡酰-β-D-葡萄糖(1),6-O-咖啡酰-D-葡萄糖(2),3-O-咖啡酰-D-葡萄糖(3),3-hydroxymethyl-2(5H)-furanone(4),β-miroside(5),cibotiumbaroside A(6),原儿茶酸(7),甘露糖(8),corchoionoside C(9),曲酸(10)。对DPPH清除能力试验结果表明,3个化合物对DPPH自由基都有较强的清除能力。结论:化合物1~5均为从狗脊中首次分离得到。化合物1,3,6均具有较强的抗氧化活性,其中化合物1的清除效果与维生素C(Vit C)相当。
OBJECTIVE: To study the chemical composition of dog ridges and explore their scavenging effects on diphenyl substituted bitter base hydrazine free radical (DPPH). Methods: The 70% ethanolic extracts of dog ’s spine were separated and purified by silica gel, gel chromatography and high performance liquid chromatography (HPLC). Their structures were identified by spectroscopic methods. The compounds were scavenged for DPPH activity test. RESULTS: Ten components were isolated from 70% ethanolic extracts of dog’s vertebra, which were 1-O-caffeoyl-β-D-glucose (3), 3-hydroxymethyl-2 (5H) -furanone (4), β-miroside (5), cibotiumbaroside A (6), protocatechuic acid (7), mannose (8), corchoionoside C (9), kojic acid (10). The DPPH scavenging ability test results show that three compounds have strong DPPH free radical scavenging capacity. Conclusion: Compounds 1 ~ 5 were isolated from the vertebrae for the first time. Compounds 1, 3 and 6 all have strong anti-oxidative activity, of which compound 1 has the same scavenging effect as vitamin C (Vit C).
