目的:探讨细胞因子IL-6在胚胎着床障碍发生发展中的作用,以及中药健胎液促进胚胎着床对子宫内膜IL-6的影响。方法:用吲哚美辛建立胚胎着床障碍的动物模型,动物分为正常对照组(正常组),胚胎着床障碍组(模型组),中药治疗组(中药组)。观察各组着床率和着床点数,用ELISA方法检测了妊娠第5天各组小鼠子宫内膜IL-6的蛋白含量,用RT-PCR方法检测妊娠第5天小鼠子宫内膜IL-6的mRNA水平。结果:模型组的胚胎着床数、着床位点数同正常组比较显著降低(P<0.01);中药组胚胎着床数、着床位点数同模型组比较明显升高(P<0.05),但仍低于正常组(P<0.05)。模型组子宫内膜IL-6蛋白量同正常组、中药组比较明显升高,中药组IL-6蛋白量同正常组比较没有显著差异(P=0.211)。模型组IL-6 mRNA表达水平同正常组、中药组比较显著升高(P<0.01),中药组IL-6 mRNA表达水平同正常组比较没有显著差异(P>0.05)。结论:中药健胎液通过调节IL-6的表达促进着床。
Objective: To investigate the role of cytokine IL-6 in the development of embryo implantation dysfunction and the effect of JST on embryo implantation on endometrial IL-6. Methods: The animal model of embryo impotence was established by indomethacin. The animals were divided into normal control group (normal group), implantation impotence group (model group) and traditional Chinese medicine treatment group (traditional Chinese medicine group). The implantation rate and implantation site of each group were observed. The content of IL-6 in endometrium was detected by ELISA method on the 5th day of pregnancy. The endometrial IL-6 -6 mRNA levels. Results: The number of embryo implantation and implantation sites in the model group was significantly lower than that in the normal group (P <0.01). The number of embryo implantation and implantation sites in the model group was significantly higher than that in the model group (P <0.05) Lower than the normal group (P <0.05). The amount of IL-6 protein in the endometrium of the model group was significantly higher than that in the normal group and the traditional Chinese medicine group, and there was no significant difference in the IL-6 protein level between the Chinese herbal medicine group and the normal group (P = 0.211). The expression level of IL-6 mRNA in the model group was significantly higher than that in the normal group and the traditional Chinese medicine group (P <0.01). The expression level of IL-6 mRNA in the Chinese herbal medicine group was not significantly different from the normal group (P> 0.05). Conclusion: JSTF can promote implantation by regulating the expression of IL-6.
