目的 :从中国姥鲨软骨组织抽提蛋白质中筛选具有新生血管抑制活性和抗肿瘤活性的分子。方法 :盐酸胍抽提姥鲨软骨蛋白 ,采用超滤和分子筛柱层析等方法分离纯化获得相对分子质量为 8.3× 10 3的蛋白质 (Sp8) ,以体外抑制血管内皮细胞增殖、体内抑制新生血管生长和抑制小鼠移植 S180肉瘤生长作为筛选指标。 结果 :Sp8可在体外抑制血管内皮细胞增殖 ,40μg/ml浓度时抑制率为 2 0 % ;在体内具有抑制新生血管生长的作用 ,兔角膜模型抑制率为 72 .7% ,鸡胚模型 (CAM)抑制率为 79% ;并能明显抑制小鼠体内移植 S180肉瘤的生长 (P<0 .0 0 1)。结论 :Sp8介导了鲨鱼软骨中的抗肿瘤活性 ,其作用可能是通过抑制肿瘤诱生新生血管形成这一途径来实现的。
Objective : To select the molecules with inhibitory activity and antitumor activity of neovascularization from the protein extracted from the Chinese shark cartilage tissue. Methods: The protein of skeletal muscle cartilage was extracted with guanidine hydrochloride. The protein with relative molecular mass of 8.3×10 3 (Sp8) was isolated and purified by ultrafiltration and molecular sieve column chromatography to inhibit the proliferation of vascular endothelial cells in vitro and inhibit the neovascularization in vivo. Growth and inhibition of mouse S180 sarcoma growth as a screening index. RESULTS: Sp8 inhibited the proliferation of vascular endothelial cells in vitro, the inhibition rate was 20% at the concentration of 40 μg/ml, and had the effect of inhibiting the growth of neovascularization in vivo. The rabbit corneal model inhibition rate was 72.7%. Chicken embryo model (CAM The inhibition rate was 79%, and it could significantly inhibit the growth of transplanted S180 sarcoma in mice (P < 0.01). Conclusion: Sp8 mediates the anti-tumor activity in shark cartilage, and its effect may be achieved by inhibiting tumor-induced neovascularization.
