目的 :建立体外培养牛眼小梁细胞 (bovinetrabecularmeshworkcell,TMcell)的方法并观察小梁细胞骨架 (cytosleleton)的形态学特征 ,尤其是actin和bubulin。方法 :采用常规方法对牛眼小梁细胞进行体外培养 ,并且免疫组织化学的方法神经原烯醇化酶及Ⅷ因子相关抗原 (NSE ,Ⅷfactorrelatedantigen) ,光学及电子透射显微镜对原代及传代培养的细胞进行形态学及生长特性的观察。并用免疫组化的方法对微丝及微管染色。结果 :牛眼小梁细胞培养成功 ,成活率 80 % - 10 0 % ,免疫组化法对actin和tubulin染色效果良好。结论 :体外培养牛眼小梁细胞技术是研究小梁细胞特性的重要实验技术。我们的研究说明选择性的收集小梁组织块是培养成功及保证成活率的有效手段。了解细胞骨架的形态学特征有助于进一步了解小梁细胞的功能
OBJECTIVE: To establish a method for culturing bovine trabecular meshwork cells (TMcell) in vitro and to observe the morphological characteristics of trabecular cytosleleton, especially actin and bubulin. METHODS: Bovine trabecular meshwork cells were cultured in vitro by conventional methods. Immunohistochemical methods including neurogenicity enolase and factor Ⅷ related antigen (NSE), optical and electron transmission microscopy were used to detect primary and subcultured cells Morphological and growth characteristics were observed. Immunohistochemistry was used to stain the microfilaments and microtubules. Results: Bovine trabecular meshwork cells were cultured successfully with the survival rate of 80% -10.0%. Immunohistochemical staining of actin and tubulin showed good results. CONCLUSION: Bovine trabecular meshwork cells cultured in vitro is an important experimental technique for studying trabecular cell characteristics. Our study shows that the selective collection of trabecular tissue mass is an effective means of successful culture and survival. Understanding the morphological features of the cytoskeleton helps to further understand the function of trabecular cells
