目的:探讨肌节同源盒基因同系物2(Msx2)和人拓扑异构酶Ⅱ-α(topoⅡ-α)在鼻内翻性乳头状瘤(SNIP)组织中的表达和意义,及其在SNIP恶变发生、发展过程中的关系。方法:采用免疫组织化学SP法分别检测Msx2和topoⅡ-α在32例SNIP,30例鼻息肉,30例恶变SNIP组织中的表达情况,其中SNIP组又根据病理形态分为轻度不典型增生组、中度不典型增生组和重度不典型增生组。结果:SNIP、恶变SNIP组织中Msx2的平均光密度值分别为0.2183±0.0598和0.2521±0.0761,均显著高于鼻息肉组织的0.1878±0.0372(P<0.05或0.01)。SNIP、恶变SNIP组织中topoⅡ-α的平均光密度分别为0.230 3±0.039 7和0.266 6±0.048 3,均显著高于鼻息肉组织的0.197 8±0.038 8(P<0.01)。Msx2和topoⅡ-α阳性表达在SNIP的3个病理形态分组间两两比较差异均有统计学意义(P<0.01或0.05),且在SNIP中的表达呈正相关(P<0.05)。结论:Msx2和topoⅡ-α在SNIP恶变的发生、发展中起重要作用,有可能成为SNIP基因治疗的靶点。
OBJECTIVE: To investigate the expression and significance of toxoid-like gene 2 (Msx2) and topoⅡ-α in nasal inverted papilloma (SNIP) SNIP malignant transformation, the development process of the relationship. Methods: The expressions of Msx2 and topoⅡ-α in 32 cases of SNIP, 30 cases of nasal polyps and 30 cases of malignant SNIP were detected by immunohistochemical SP method. The SNIP group was divided into mild dysplasia group , Moderate dysplasia group and severe dysplasia group. Results: The average optical density of Msx2 in SNIP and malignant SNIP tissues were 0.2183 ± 0.0598 and 0.2521 ± 0.0761, respectively, which were significantly higher than those in nasal polyps (0.1878 ± 0.0372, P <0.05 or 0.01). The average optical density of topoⅡ-α in SNIP and malignant SNIP tissues were 0.230 3 ± 0.039 7 and 0.266 6 ± 0.048 3, respectively, which were significantly higher than that of nasal polyps (0.197 8 ± 0.038 8, P <0.01). The positive expression rates of Msx2 and topoⅡ-α were statistically significant (P <0.01 or 0.05) among the three SNIP pathological groups, and there was a positive correlation between Msx2 and topoⅡ-α in SNIP (P <0.05). Conclusion: Msx2 and topoⅡ-α play an important role in the occurrence and development of SNIP malignancies, which may be the target of SNIP gene therapy.
