脱水响应元件结合蛋白(DREB)转录因子在应激反应相关基因的调控中起重要作用。本研究利用RT-PCR方法从梭梭组织中获得Ha DREB2A基因开放阅读框(ORF)全长(Gen Bank登录号为:KP765243)。生物信息学分析表明,该基因ORF长1 083 bp,编码360个氨基酸,蛋白的分子量为40.5 k D,理论等电点(p I)为5.07,该蛋白在90(K)~143(R)区域含有一个典型的AP2保守结构域。通过物种间系统发育树可以看出Ha DREB2A与Sb DREB2A进化关系最接近,二者属于同一进化分支。半定量RT-PCR表达模式分析显示,该基因在梭梭同化枝、根、茎、花和种子中均有表达,且在同化枝中的表达量最高,并且受干旱、高盐和外源ABA的诱导。
Dehydration response element binding protein (DREB) transcription factor plays an important role in the regulation of stress related genes. In this study, the full-length ORF of Ha DREB2A gene (GenBank accession number: KP765243) was obtained from Haloxylon ammodendron by RT-PCR. Bioinformatics analysis showed that the ORF of the gene was 1 083 bp encoding a protein of 360 amino acids with a molecular weight of 40.5 kD and a theoretical isoelectric point (pI) of 5.07. The ORF of the gene was between 90 (K) and 143 (R) The region contains a typical AP2 conserved domain. Through the phylogenetic tree between species, it can be seen that the evolutionary relationship between Ha DREB2A and Sb DREB2A is the closest, both belong to the same evolutionary branch. Semi-quantitative RT-PCR analysis showed that the gene was expressed in the assimilate branches, roots, stems, flowers and seeds of Haloxylon ammodendron, and its expression level was the highest in the assimilating branches. And it was affected by drought, high salt and exogenous ABA Induction.
