目的 :探讨bFGF表达及FGFR1基因突变与涎腺肿瘤发生发展的关系。方法 :采用免疫组织化学技术和RT -PCR -SSCP法 ,对 14例多形性腺瘤、9例腺样囊性癌组织和 11例正常涎腺组织中bFGF表达及相应组织内FGFR1胞内段结构基因进行了检测。结果 :bFGF在多形性腺瘤、腺样囊性癌中的表达明显高于正常涎腺组织 ;FGFR1在 14例多形性腺瘤中 4例检出基因突变 ,9例腺样囊性癌组织中 2例检出突变 ,且发现突变基因位于FGFR1胞内段的近膜区及部分激酶I区。结论 :FGFR1细胞内段激酶活性区的突变可能造成bFGF /FGFR1信号传递异常 ,进而参与延腺肿瘤的发生发展
Objective: To investigate the relationship between the expression of bFGF and the mutation of FGFR1 and the development of salivary gland tumors. Methods: The expression of bFGF in 14 cases of pleomorphic adenoma, 9 cases of adenoid cystic carcinoma and 11 cases of normal salivary gland tissues were detected by immunohistochemistry and RT-PCR-SDS-PCR method. The intracellular FGFR1 expression in the corresponding tissues Genes were tested. Results: The expression of bFGF in pleomorphic adenoma and adenoid cystic carcinoma was significantly higher than that in normal salivary gland. FGFR1 gene mutations were detected in 4 of 14 pleomorphic adenomas and in 9 adenoid cystic carcinomas Two cases were detected mutations, and found that the mutant gene located in the intracellular FGFR1 proximal membrane area and part of the kinase I region. Conclusion: Mutations in the kinase domain of FGFR1 may result in the abnormal signal transduction of bFGF / FGFR1, which may be involved in the development of the tumor
