目的观察基质金属蛋白酶-9(MMP-9)在高蛋氨酸喂养所致的高同型半胱氨酸血症大鼠中的表达及意义。方法健康wistar大鼠20只,随机分为:正常对照组、高蛋氨酸组,每组10只,并分别给予普通饲料,普通饲料中添加蛋氨酸(40g/kg饲料)。8周后,腹主动脉采血,同时摘取两侧颈总动脉。采用ELISA法检测大鼠血浆总同型半胱氨酸浓度与MMP-9表达;采用逆转录多聚酶链式反应(RT-PCR)及HE染色分别观察动脉组织中MMP-9mRNA的表达及动脉损伤情况。结果高蛋氨酸组中血浆同型半胱氨酸浓度,血浆MMP-9蛋白浓度及动脉组织中MMP-9mRNA的表达水平均显著高于正常对照组,差异具有统计学意义(p<0.01);相关性分析显示同型半胱氨酸浓度与MMP-9的表达存在线性正相关关系。同时镜下观察药物高蛋氨酸组动脉损伤程度较对照组明显。结论高同型半胱氨酸可通过上调MMP-9的表达发挥其致动脉粥样硬化的作用。
Objective To investigate the expression and significance of matrix metalloproteinase-9 (MMP-9) in hyperhomocysteinemia rats induced by high methionine. Methods Twenty healthy wistar rats were randomly divided into normal control group and high methionine group, with 10 mice in each group. The animals were given normal diet and methionine (40g / kg diet) respectively. After 8 weeks, abdominal aorta blood, while removing the common carotid artery on both sides. The concentration of plasma homocysteine and the expression of MMP-9 in plasma were detected by ELISA. The expression of MMP-9 mRNA and arterial injury were observed by reverse transcription-polymerase chain reaction (RT-PCR) and HE staining. Results The levels of plasma homocysteine, the concentration of plasma MMP-9 and the level of MMP-9 mRNA in arterial tissue in high methionine group were significantly higher than those in normal control group (p <0.01), and the correlation was significant Analysis showed that homocysteine concentration and MMP-9 expression there is a linear positive correlation. At the same time, the degree of arterial injury in the high-methionine group was significantly higher than that in the control group. Conclusion Homocysteine can exert its effect on atherosclerosis by up-regulating the expression of MMP-9.
