目的 :活性维生素D3 (1,2 5 (OH) 2 D3 )是治疗骨质疏松调整钙磷代谢的常用药物 ,本研究试图研究它对动脉钙化发展过程中的影响。方法 :体外培养牛主动脉平滑肌细胞 ,建立体外血管钙化模型 ,在培养介质中加入 1,2 5(OH) 2 D3 ,观察细胞钙沉积及与之相关的细胞碱性磷酸酶活性和骨钙素的变化情况 ;结果 :10 -9mol/L的 1,2 5(OH) 2 D3 培养 10d后 ,钙化细胞的钙沉积增加了 16 .2 5 % ,细胞内碱性磷酸酶活性增加了 30 1% ,骨钙素分泌增加了 5 8.3% ,提示 1,2 5 (OH) 2 D3 可以加重钙化培养的血管平滑肌细胞的钙盐沉积 ,而对作为对照的普通血管平滑肌细胞此种作用不明显。结论 :1,2 5 (OH) 2 D3 可促进钙化培养的血管平滑肌细胞的钙质沉积 ,其作用可能与增加细胞碱性磷酸酶活化及促进细胞向成骨细胞转化有关
PURPOSE: Active vitamin D3 (1,25 (OH) 2 D3) is a commonly used drug for the treatment of osteoporosis to regulate calcium and phosphorus metabolism. This study attempted to study its effect on the development of arterial calcification. Methods: Bovine aortic smooth muscle cells were cultured in vitro and the in vitro vascular calcification model was established. 1,25 (OH) 2 D3 was added to the culture medium to observe the cell calcium deposition and the related cell alkaline phosphatase activity and osteocalcin Results: The calcium deposition of calcified cells increased by 16.25% after 10-9 mol / L 1,25 (OH) 2 D3 was cultured for 10 days, and the activity of alkaline phosphatase increased by 30 1% , Osteocalcin increased by 8.33%, suggesting that 1,25 (OH) 2 D3 can increase the calcification of cultured vascular smooth muscle cells of calcium deposition, while as a control of ordinary vascular smooth muscle cells this effect is not obvious. CONCLUSION: 1,25 (OH) 2 D3 can promote the calcification of cultured vascular smooth muscle cells, which may be related to the increase of cell alkaline phosphatase activation and the promotion of cell transformation into osteoblasts
