应用人肥胖蛋白(ObeseproteinOP57-92活性肽段)及其特异性兔抗人OP(57-92)血清建立了液相竞争放射免疫分析。标准曲线范围为20~1280pg/管,灵敏度为5pg/管;批内、批间变异系数分别为8.26%和10.56%;回收率为72.9~101.2%、平均为84;7%;OP-Ab与相关蛋白及多肽如人血白蛋白、甲状腺素(T3、T4)、胰岛素、胰高血糖素、促甲状腺素及心钠素等无明显交叉反应。肥胖成人、肥胖小儿和糖尿病患者血浆OP测定值显著低于健康非肥胖对照组(P<0.01)。结果提示,OP对调节体重及内分泌代谢起重要作用,循环OP含量不足可能是肥胖及糖尿病发病的重要因素之一。人血浆OP放免的建立,对肥胖和糖尿病的临床诊断与基础研究提供了一种新的手段。
Liquid-phase competitive radioimmunoassay was established using human Obese protein P57-92 active peptide and its specific rabbit anti-human OP (57-92) serum. The standard curve ranged from 20 to 1280pg / tube, with a sensitivity of 5pg / tube. The coefficient of variation (CV) was 8.26% and 10.56%, respectively. The average recoveries ranged from 72.9% to 101.2% with an average of 84 ; 7%; OP-Ab did not significantly cross-react with related proteins and peptides such as human serum albumin, thyroxine (T3, T4), insulin, glucagon, thyrotropin and atrial natriuretic peptide. The measured values of plasma OP in obese adults, obese children and diabetic patients were significantly lower than those in healthy non-obese controls (P <0.01). The results suggest that OP plays an important role in the regulation of body weight and endocrine metabolism. The lack of circulating OP may be one of the important factors of obesity and diabetes. The establishment of human plasma OP radioimmunoassay provides a new means of clinical diagnosis and basic research on obesity and diabetes.
