目的探讨检测慢性淋巴细胞白血病患者孕激素受体B基因CpG岛甲基化状态改变的临床意义。方法提取外周血DNA,经亚硫酸盐修饰,然后采用甲基化特异性PCR技术检测9例慢性淋巴细胞白血病患者及5例正常对照外周血DNA孕激素受体B基因CpG岛甲基化状态。结果在9例慢性淋巴细胞白血病患者外周血中5例成功地检测到孕激素受体B基因甲基化异常,而正常对照均没有发现甲基化异常。结论对慢淋孕激素受体B基因异常甲基化的检测该方法可能对监测肿瘤进程有一定作用。
Objective To investigate the clinical significance of detecting methylation status of CpG island of progesterone receptor B gene in patients with chronic lymphocytic leukemia. Methods Peripheral blood DNA was extracted and sulfite-modified. Methylation-specific PCR was used to detect the methylation status of CpG island of the DNA progesterone receptor B gene in 9 patients with chronic lymphocytic leukemia and 5 normal controls. Results The methylation abnormalities of progesterone receptor B gene were successfully detected in peripheral blood of 9 patients with chronic lymphocytic leukemia and no methylation abnormalities were found in normal controls. Conclusion The detection of abnormal methylation of progesterone receptor B gene may play a role in the monitoring of tumor progression.
