已知IL-12、IL-7可对T细胞、NK细胞产生多种生物学效应,但有关IL-12、IL-7和IL-2间的相互作用至今尚未明确.本文应用同位素3~H-TdR掺入法和3~H-TdR释放法比较了重组IL-12、IL-7、IL-2单独或协同对健康人外周血单个核细胞(PBMC)的体外增殖和细胞毒活性的影响,结果表明,IL-7单独即可促进PBMC增殖分化和诱导产生细胞毒活性,与IL-2协同可使二种作用均增强,并高于单独用IL-2时的效应.IL-12单独不能引起PBMC增殖,但可诱导PBMC产生细胞毒活性,与IL-2协同可使这种效应明显增强.IL-12与IL-7协同作用于PBMC引起的增殖反应呈现一种剂量依赖方式.
It is known that IL-12 and IL-7 can produce a variety of biological effects on T cells and NK cells, but the interactions between IL-12, IL-7 and IL-2 have not yet been clarified. -TdR incorporation method and 3~H-TdR release method were used to compare the effects of recombinant IL-12, IL-7 and IL-2 alone or synergistically on the proliferation and cytotoxicity of peripheral blood mononuclear cells (PBMC) in healthy volunteers. The results show that IL-7 alone can promote the proliferation and differentiation of PBMCs and induce cytotoxic activity. The synergistic effect with IL-2 can enhance both effects and is higher than that of IL-2 alone. IL-12 alone It can’t induce the proliferation of PBMC, but it can induce the cytotoxic activity of PBMC, and this effect can be obviously enhanced by the cooperation with IL-2. The proliferative response of IL-12 and IL-7 synergistically induced by PBMC presents a dose-dependent manner.
