本文报道以MTT法检测LAK细胞冻融液对肿瘤细胞直接杀伤作用,确立检测过程中的某些最适条件。发现以K562细胞和NKM-45细胞为靶细胞时,试验最适细胞浓度分别为2×106/ml和1.5×106/ml。为阻断检测过程中肿瘤细胞增殖对结果的影响,需加入终浓度为lug/ml的放线菌素D。实验结果提示,LAK细胞可能通过释放某些杀伤分子介导对肿瘤细胞的直接杀伤作用。
This paper reports the direct killing effect of freeze-thaw fluid from LAK cells on tumor cells by MTT assay and establishes some of the optimal conditions for the detection process. When K562 cells and NKM-45 cells were used as target cells, the optimal cell concentrations for the assay were 2×10 6 /ml and 1.5×10 6 /ml, respectively. In order to block the effect of tumor cell proliferation on the results during the assay, actinomycin D was added at a final concentration of lug/ml. The experimental results suggest that LAK cells may mediate direct killing of tumor cells by releasing certain killer molecules.
