作者采用~(125)I标记的人乳铁蛋白(hLf)结合试验,对属于24种不同血清型的63株肺炎链球菌进行检测。以结合率<5%为阴性,结果有88%的受试菌株呈阳性,其中结合率>30%的占43%。从竞争性抑制结合试验发现,与hLf结合的细菌表面组分有两种,其解离常数分别为5.7×10~(-8)和2.74×10~(-7)M。以未标记牛Lf或人转铁蛋白作为竞争物,均不影响hLf的特异性结合。以胰蛋白酶或链霉蛋白酶E处理肺炎链球菌,则其原有的hLf结合活性全部丧失。
The authors tested 63 strains of S. pneumoniae belonging to 24 different serotypes using the ~ (125) I labeled human lactoferrin (hLf) binding assay. At a binding rate of <5%, 88% of the tested strains were positive, with a binding rate of> 30% accounting for 43%. From the competitive inhibition binding assay, it was found that there are two bacterial surface components that bind to hLf with dissociation constants of 5.7 × 10 -8 and 2.74 × 10 -7 M, respectively. Unlabeled bovine Lf or human transferrin as a competitor did not affect the specific binding of hLf. Treatment of Streptococcus pneumoniae with trypsin or pronase E lost its original hLf binding activity.
