目的研究中药髓复康对脊髓损伤后,背根节(DRG)神经元降钙素基因相关肽(CGRP)表达的影响。探讨髓复康效应的机理。方法造成下胸髓半横断损伤的Wistar大鼠模型,用免疫细胞化学法和定量图像分析法检查CGRP表达的变化。结果①脊髓半横断引起同侧首、尾侧各3个DRG内,CGRP表达的明显和持续性下调;②损伤后给服髓复康冲剂,可以明显减轻,但不能完全阻止,脊髓损伤诱发的CGRP表达下调;③损伤后给服中药补阳还五汤(或用大剂量的氢化考地松)治疗,不能减轻脊髓损伤诱发的CGRP表达下调;④脊髓损伤后服用髓复康的大鼠,其脊髓损伤区有明显的修复再生。结论结果不仅表明CGRP表达的变化与脊髓组织的损伤和再生有一定的关联;而且也表明,髓复康通过对CGRPmRNA表达的调制,影响神经元DNA的合成,可能是它促进脊髓损伤组织的修复再生的机理之一。此假设还需进一步确认。
OBJECTIVE: To study the effect of traditional Chinese medicine Puyeukang on the expression of calcitonin gene-related peptide (CGRP) in dorsal root ganglion (DRG) neurons after spinal cord injury. Explore the mechanism of the piroxicam effect. Methods The Wistar rat model of semi-transverse thoracic spinal cord injury was established. CGRP expression was examined by immunocytochemistry and quantitative image analysis. Results 1 Semi-transected spinal cord resulted in obvious and persistent down-regulation of CGRP expression in 3 DRGs in the ipsilateral and caudal ipsilateral. 2 Post-injury Gifu Fukang Granules could be significantly relieved, but could not be completely prevented. Spinal Cord Injury Induced The expression of CGRP was down-regulated; 3 After the injury, taking Chinese medicine Buyang Huanwu Decoction (or high-dose hydrocortisone) did not reduce the down-regulation of CGRP induced by spinal cord injury; 4 The rats taking spinal cord injury after spinal cord injury, The spinal cord injury area has obvious repair and regeneration. Conclusions The results not only indicate that changes in CGRP expression are associated with spinal cord tissue injury and regeneration; but also indicate that the treatment of pharmacological effects of piroxicam on the expression of CGRP mRNA affects the synthesis of neuronal DNA, possibly because it promotes spinal cord injury.
