原子力显微术由于其独特的成像方式,为生物医学领域高分辨表征与成像研究提供了崭新的手段,但其在活细胞及新鲜组织成像上仍存在着巨大挑战.本文结合作者在活细胞及新鲜组织成像方面的工作,对原子力显微术两种常用工作模式(接触模式和轻敲模式)的特点和局限、新的轻敲模式(磁驱动轻敲模式,MAC mode)用于活体生物样本的优势以及MAC mode所提供的一种反映样品表面新信息的TREC像进行了叙述,进而介绍了目前原子力显微术在活体生物样本成像方法及表征研究方面的进展.
Because of its unique imaging method, atomic force microscopy provides a new method for high-resolution characterization and imaging in the field of biomedicine, but there are still great challenges in the imaging of living cells and fresh tissues. Fresh Tissue Imaging, the new tapping mode (MAC mode) is used in living biological samples for the features and limitations of two common working modes of atomic force microscopy (contact mode and tapping mode) The advantages of MAC mode and the TREC image provided by MAC mode, which reflect the new information on the surface of the sample, are described. Then the progress of the present methods and the characterization of the biological samples by AFM is introduced.
