研究建立经萃取后衍生化高效液相色谱法,用于测定碘化油顺铂乳剂中顺铂含量。方法:乳剂中顺铂由二甲基亚砜(DMSO)萃取后,用二乙基硫代氨基甲酸钠(DDTC)衍生化,其产物可在254nm波长处被检出,以氯化镍为内标物,固定相为UBondapakC18H37化学键合反相固定相,流动相为甲醇∶水(80∶20),流速为1.0ml·min-1。结果:测得平均萃取回收率为78.4%,RSD<5%。结论:应用本法测定乳剂中顺铂含量操作简单,快速。
The establishment of the extraction and derivatization of high performance liquid chromatography for the determination of cisplatin iodized oil emulsion cisplatin content. METHODS: Cisplatin in emulsion was extracted with dimethyl sulfoxide (DMSO) and derivatized with sodium diethylcarbamaminate (DDTC). The product was detected at 254 nm and nickel chloride was used as internal standard The stationary phase was UBondapakC18H37 chemically bonded reversed-phase stationary phase. The mobile phase consisted of methanol: water (80:20) and the flow rate was 1.0ml · min-1. Results: The average extraction recovery was 78.4% with RSD <5%. Conclusion: The determination of cisplatin content in the emulsion by this method is simple and fast.
