目的:探讨HuR siRNA对人肺腺癌A549株细胞增殖、迁移及侵袭的影响及其可能机制。方法:HuR siRNA瞬时转染肺腺癌A549细胞24 h后,CCK-8实验、集落形成实验检测细胞的增殖能力,划痕实验检测细胞迁移能力,Transwell实验检测细胞侵袭能力,qPCR和Western blotting检测HuR及基质金属蛋白酶(MMP)-9基因mRNA和蛋白质表达。结果:HuR siRNA能够抑制人肺腺癌A549细胞的增殖、迁移及侵袭;HuR siRNA显著降低A549细胞HuR、MMP-9 mRNA和蛋白表达(P<0.01)。结论:下调HuR能抑制肺腺癌A549细胞增殖、迁移及侵袭,其机制与下调MMP-9有关。
Objective: To investigate the effect of HuR siRNA on the proliferation, migration and invasion of human lung adenocarcinoma A549 cells and its possible mechanism. METHODS: Human lung adenocarcinoma A549 cells were transiently transfected with HuR siRNA for 24 h. CCK-8 assay and colony formation assay were used to detect the proliferation of cells. Scratch assay was used to detect cell migration. Transwell assay was used to detect cell invasion ability. QPCR and Western blotting HuR and matrix metalloproteinase (MMP) -9 gene mRNA and protein expression. Results: HuR siRNA could inhibit the proliferation, migration and invasion of human lung adenocarcinoma A549 cells. HuR siRNA significantly decreased the expression of HuR and MMP-9 mRNA and protein in A549 cells (P <0.01). Conclusion: Down-regulation of HuR can inhibit the proliferation, migration and invasion of lung adenocarcinoma A549 cells. The mechanism is related to the down-regulation of MMP-9.
