目的:制备超抗原(SAg)葡萄球菌肠毒素A(SEA)与抗人肝细胞癌单克隆抗体(McAb)HAb18F(ab′)2段的结合物,并对其活性进行鉴定.方法:提取MCAbHAb18并用木瓜蛋白酶切取其F(ab′)2段,用SPDP制备HAb18F(ab′)2-SEA结合物,凝胶色谱柱纯化后用SDS-PAGE鉴定各收集峰,免疫组化ABC法鉴定结合物中的HAb18F(ab′)2的抗体活性,MMT法鉴定结合物中的SEA活化T细胞的活性.结果:成功地制备了HAb18F(ab′)2-SEA结合物,并且制备的结合物具有肝癌抗体活性和活化外周血单个核细胞的功能.结论:SPDP是一种很好的蛋白质交联剂,本实验制备的HAb18F(ab′)2-SEA结合物可用于McAb与SAg结合物导向治疗肝癌的实验研究.
OBJECTIVE: To prepare a conjugate of super antigen (SAg) staphylococcal enterotoxin A (SEA) and anti-human hepatocellular carcinoma monoclonal antibody (McAb) HAb18F (ab’) 2 and to identify its activity. METHODS: MCAbHAb18 was extracted and its F(ab’)2 fragment was digested with papain. The HAb18F(ab’)2-SEA conjugate was prepared using SPDP. After purification by gel column chromatography, the collected peaks were identified by SDS-PAGE and immunohistochemical ABC. The antibody activity of HAb18F(ab’)2 in the conjugate was determined and the activity of SEA activated T cells in the conjugate was identified by MMT assay. RESULTS: HAb18F(ab’)2-SEA conjugate was successfully prepared, and the prepared conjugate had the activity of liver cancer antibody and the function of activated peripheral blood mononuclear cells. Conclusion: SPDP is a good protein cross-linker. The HAb18F(ab′)2-SEA conjugate prepared in this experiment can be used for the experimental study of McAb and SAg conjugate-guided treatment of liver cancer.
