目的:研究化瘀通孕方对输卵管炎性阻塞性不孕症大鼠输卵管细胞间黏附分子-1(ICAM-1)表达及血流变学的影响,从分子生物学角度探讨化瘀通孕方的作用机制。方法:SPF级Wistar雌性大鼠70只,随机选取60只大鼠采用混合菌法制作输卵管炎性不孕症大鼠模型,剩余10只作为空白组。选择造模成功的大鼠50只,随机分为模型组、西药组、化瘀通孕方低、中、高各剂量组,各10只。化瘀通孕方低、中、高各剂量组分别按6.09、12.18、24.36 g/(kg·d)灌胃化瘀通孕方,西药组灌胃头孢呋辛酯片52.5 mg/(kg·d)和甲硝唑片63 mg/(kg·d),空白组和模型组予等容量生理盐水灌胃。检测大鼠血液流变学指标和输卵管组织ICAM-1蛋白的表达。结果:空白组大鼠输卵管不表达ICAM-1。与空白组比较,模型组大鼠全血黏度高切和低切、红细胞刚性指数、红细胞聚集指数、红细胞压积均显著升高(P<0.01),说明造模成功。与模型组比较,西药组和化瘀通孕方低、中、高各剂量组大鼠输卵管ICAM-1蛋白表达累积面积和平均光密度均显著减小,全血黏度高切和低切、红细胞刚性指数、红细胞聚集指数、红细胞压积均显著降低,差异均有统计学意义(P<0.01)。与西药组比较,化瘀通孕方低剂量组大鼠输卵管ICAM-1蛋白表达累积面积较高;化瘀通孕方中、高剂量组大鼠输卵管ICAM-1蛋白表达累积面积、大鼠全血黏度高切和低切、红细胞刚性指数、红细胞压积均显著降低,差异均有统计学意义(P<0.05)。与化瘀通孕方高剂量组比较,化瘀通孕方低、中剂量组大鼠输卵管ICAM-1蛋白表达累积面积和平均光密度均显著较高,全血黏度高切和低切、红细胞刚性指数、红细胞聚集指数、红细胞压积均显著较高,差异均有统计学意义(P<0.05)。结论:化瘀通孕方能明显改善大鼠血液流变学指标,并可以降低输卵管ICAM-1蛋白的表达,为临床治疗输卵管炎性阻塞性不孕症提供可靠的实验依据。
Objective: To study the effect of Huayu Tong-Jun Decoction on the expression of tubal intercellular adhesion molecule-1 (ICAM-1) and the hemorheology in tubal obstruction infertility rats. To explore the effect of Huayu Tong pregnant Party’s mechanism of action. Methods: Seventy SPF Wistar female rats were randomly selected. Sixty rats were randomly selected to make a rat model of tubal infertility with mixed bacteria. The remaining 10 rabbits were used as a blank group. Fifty rats were selected and randomly divided into model group, western medicine group and low, medium and high dose of Huayu Tong pregnant, each of 10 rats. Huayu Tong pregnant square low, middle and high doses of each group were 6.09,12.18,24.36 g / (kg · d) gavage Huayu Tong pregnant side, Western medicine group cefuroxime axetil tablets 52.5 mg / (kg · d) and metronidazole tablets 63 mg / (kg · d), the blank group and the model group were given equal volume of normal saline. The hemorrheological indexes and the expression of ICAM-1 protein in oviduct tissue were detected. Results: The fallopian tube of the blank group did not express ICAM-1. Compared with the blank group, whole blood viscosity in the model group was significantly higher than that in the control group (P <0.01), indicating that the modeling was successful. Compared with the model group, the cumulative area and average optical density of ICAM-1 protein expression in the fallopian tube, the middle and high dose groups in the WM group and the Huayu Tongji decoction group were significantly decreased, the whole blood viscosity was high and low cut, Rigid index, erythrocyte aggregation index and hematocrit were significantly decreased, the differences were statistically significant (P <0.01). Compared with the western medicine group, the accumulative area of ICAM-1 protein expression in the fallopian tube in the low-dose Huayu Tong-pregnant decoction group was higher than that in the Huayu Tong-pregnant decoction group. The ICAM- High and low shear viscosity of blood viscosity, erythrocyte rigidity index and hematocrit were significantly decreased, the difference was statistically significant (P <0.05). Compared with the high-dose Huayu Tong decoction group, the accumulative area and the average optical density of ICAM-1 protein expression in the tubal tissues of Huayu Tong pregnant decoction group were significantly higher, the whole blood viscosity was high and low cut, Rigid index, erythrocyte aggregation index, hematocrit were significantly higher, the differences were statistically significant (P <0.05). Conclusion: Huayu Tongsu Decoction can significantly improve the hemorheology indexes of rats and reduce the expression of ICAM-1 protein in the fallopian tubes, which can provide a reliable experimental basis for clinical treatment of tubal obstructive infertility.
