目的和方法:实验以培养的人脐静脉内皮细胞(HUVEC)为研究对象,应用大剂量肿瘤坏死因子(TNF)造成HUVEC损伤,观察培养上清液血管紧张素转换酶(ACE)及HUVEC的超微结构变化,研究蜕皮甾酮(EDS)对内皮细胞损伤的影响。结果:大剂量TNF作用于HUVEC时,其培养上清液的ACE非常明显增高,超微结构破坏;应用EDS,则培养上清液的ACE有所减少、超微结构的损害程度有所减轻。结论:EDS能够减轻人血管内皮细胞的损伤,揭示了EDS在抗血管内皮细胞损伤方面的价值。
PURPOSE AND METHODS: The cultured human umbilical vein endothelial cells (HUVECs) were used as experimental subjects. High dose of tumor necrosis factor (TNF) was used to induce HUVEC injury. The effects of supernatant of angiotensin converting enzyme (ACE) Micro-structure changes, the study of ecdysterone (EDS) on the endothelial cell injury. Results: When high dose of TNF was applied to HUVEC, the ACE in culture supernatant was significantly increased and the ultrastructure was destroyed. With EDS, the ACE in culture supernatant was reduced and the ultrastructure damage was alleviated. Conclusion: EDS can reduce the damage of human vascular endothelial cells and reveal the value of EDS in anti-vascular endothelial cell injury.
