目的:改进附骨疽动物模型的制备方法并完善模型验证指标。方法:将新西兰兔随机分为空白对照组、附骨疽模型组与细菌感染模型组,附骨疽模型组动物以冰敷(10min/d)15d,采用金黄色葡萄球菌局部感染方法造模,细菌感染模型组以金黄色葡萄球菌局部感染方法造模。以血液流变学指标、血液炎性反应指标、病理组织学指标验证模型。结果:附骨疽模型组与空白对照组比较,血液黏度增高(P<0.01),附骨疽模型组与细菌感染模型组比较,白细胞计数、超敏C反应蛋白显著升高(P<0.01);组织病理学检查显示:附骨疽模型组坏死组织范围较大,肉芽组织显著增多。结论:局部冰敷结合细菌感染的方法可用于附骨疽模型的制备,为附骨疽病因探究及中药疗效评价提供方法与依据。
OBJECTIVE: To improve the preparation method of animal model of appendicitis and to improve the model verification index. Methods: New Zealand white rabbits were randomly divided into blank control group, model group with bacterial adhesion and model group with bacterial infection. The animals with model group were treated with ice stabbing (10 min / d) for 15 days by local infection with Staphylococcus aureus. Bacterial infection model group Staphylococcus aureus local infection method modeling. Hemorheology index, blood inflammatory response index and histopathological index validation model. Results: Compared with the blank control group, the blood viscosity increased (P <0.01), and the leukocyte count and the high sensitive C reactive protein in the model group were significantly higher than those in the bacterial infection model group (P <0.01) Histopathological examination showed that there was a large range of necrotic tissue in the model group and a significant increase in granulation tissue. CONCLUSION: The method of local ice pack combined with bacterial infection can be used for the preparation of the model of epididymal ganglion, and provides the method and basis for the exploration of the causal factor of appendicitis and evaluation of the curative effect of traditional Chinese medicine.
