将白花泡桐(Paulowniafortunei)丛生芽转至于MS+IBA1.5mg/L培养基上,6d丛生芽长出不定根.用液相色谱法分析白花泡桐生根过程中内源IAA和CTK的变化,结果表明:在生根过程中,第2dIAA含量达最高峰(11.11μg.g ̄(-1)FW),然后下降,时至第6d为最低(3.70μg·g ̄(-1)FW).cTK第2d出现一个峰(1.36μg.g ̄(-1)FW)后逐步下降,到第4d降至最低值(o.66μg·g ̄(-1)FW)但过后又上升,至第6d不定根出现时峰值达(1.78μg.g ̄(-1)FW).而CTK/IAA的比值则是以第2d为最低(0.12),然后开始迅速上升,至第6d不定根出现时,其比值达0.48.核酸含量测定表明:DNA的含量在生根过程中逐步下降,当不定根出现时才开始回升.RNA的含量则是前期下降,随后迅速升高.根分化不仅与IAA和CTK含量有关,而且与两者的比值变化关系更密切.
The cluster shoots of Paulownia fortunei were transferred to MS + IBA1.5mg / L medium, and the adventitious roots grew on the 6th day. The changes of endogenous IAA and CTK during the rooting process of Paulownia involucrata were analyzed by liquid chromatography. The results showed that the content of IAA and CTK reached the peak (11.11μg.g ~ (-1) FW) on the 2nd day after rooting, then decreased, From the 6th day to the lowest (3.70μg · g -1 FW). A peak of cTK appeared on day 2d (1.36μg.g ~ (-1) FW) and then decreased to its lowest value (0.66μg · g ~ (-1) FW) on day 4, 6d adventitious roots appeared up to the peak (1.78μg.g ~ (-1) FW). However, the ratio of CTK / IAA was the lowest (0.12) at the 2nd day and then began to increase rapidly. When the adventitious roots appeared on the 6th day, the ratio reached 0.48. The determination of nucleic acid content showed that the content of DNA decreased gradually during rooting and began to rise when adventitious roots appeared. The content of RNA was decreased in the early period and then rapidly increased. Root differentiation is not only related to the content of IAA and CTK, but also to the ratio of the two.
