目的分析骨肉瘤和骨软骨瘤患者血清蛋白指纹图谱的差异,筛选对骨肉瘤早期诊断有意义的特异性蛋白。方法收集河南省洛阳正骨医院2007年6月至2010年1月病理确诊的骨肉瘤25例、单发骨软骨瘤16例和性别、年龄与上两组患者匹配的26例健康志愿者,采用SELDI-TOF-MS质谱仪方法对这67例静脉血液离心获得的血清进行蛋白指纹图谱分析。结果我们发现2个特异性蛋白质荷比(M/Z)具有差异性表达,即3954Da、6438Da。在学习模式下其诊断灵敏度96.0%,特异度96.1%,在测试模式下其诊断灵敏度98.5%,特异度100.0%,采用leave-one-out互证交叉检验验证,认为该决策树盲法筛选分组的正确率可达67/67=100%,灵敏度为25/25=100%;特异性为42/42=100%。结论采用SELDI-TOF-MS方法筛选出的2个特异性蛋白可能对骨肉瘤的早期诊断和治疗监测有临床参考价值。
Objective To analyze the difference of serum protein fingerprints between patients with osteosarcoma and osteochondroma and to screen the specific proteins that are of significance in the early diagnosis of osteosarcoma. Methods Twenty-five patients with osteosarcoma, 16 patients with osteochondroma and 26 healthy volunteers matched for sex and age were collected from June 2007 to January 2010 in Luoyang Orthopedics Hospital of Henan Province. SELDI -TOF-MS mass spectrometry of these 67 cases of venous blood serum obtained by protein fingerprinting analysis. As a result, we found that the two specific protein charge ratios (M / Z) were differentially expressed, that is, 3954 Da and 6438 Da. Its diagnostic sensitivity is 96.0% and its specificity is 96.1% in study mode. Its diagnostic sensitivity is 98.5% and its specificity is 100.0% in test mode.
