目的预测人二氨氧化酶 ( DAO)氨基酸序列片段 ( aa No.5 2 4- 6 18)的 B-细胞表位。方法应用 6种参数和方法进行综合分析 ,包括 Hopp & Woods亲水性参数、抗原性参数、可及性参数、β-转角、万氏及吴氏综合预测方法。结果显示 B-细胞识别的表位可能在 5 43- 5 5 3和 5 6 9- 5 95残基或其附近 ,5 6 9- 5 95残基 4种参数和方法的预测值均高于 5 43- 5 5 3残基 ,这两个被预测的表位均含有β-转角和不规则卷曲结构。结论本研究为应用合成肽抗原制备抗人 DAO抗体提供了依据
Objective To predict the B-cell epitopes of the amino acid sequence of human diammine oxidase (DAO) (aa No.5 2 4- 6 18). Methods Six kinds of parameters and methods were used for comprehensive analysis, including Hopp & Woods hydrophilicity parameters, antigenicity parameters, accessibility parameters, β-turn angle, Wanhs and Wu’s comprehensive prediction methods. The results showed that the predicted B-cell epitopes may be at or near 5 43-5 3 and 5 6 9-5 5 95 residues, and the predicted values of the 4 parameters and methods of the 5 6 9-5 5 residues are higher than 5 43-535 residues, both of which predicted epitopes contain beta-turns and irregularly curled structures. Conclusion This study provides a basis for the preparation of anti-human DAO antibodies using synthetic peptide antigens
