为观察人单纯疱疹Ⅱ型病毒感染对培养的牛主动脉平滑机细胞形态及合成胶原的影响,用50%组织培养感染量的病毒悬液于37℃感染培养的牛主动脉平滑肌细胞,于倒置显微镜和电镜下观察平滑肌细胞形态改变,以3H-脯氨酸掺入平滑肌细胞合成的3H-羟脯氨酸的量作为测定胶原的指标。结果发现,感染24h后,平滑肌细胞逐渐出现细胞变圆、肿胀和局部坏死脱落等形态改变;感染24、36和48h后平滑肌细胞胶原的含量与对照组相比明显降低。提示人单纯疱疹Ⅱ型病毒可能作为一种局部损害因子,抑制血管平滑肌细胞胶原合成,损害中滑肌细胞,可参与动脉粥样硬化的发病过程。
To observe the influence of human herpes simplex virus type 2 infection on the cell morphology and collagen synthesis of cultured bovine aortic smooth muscle cells, the infected bovine aortic smooth muscle cells were infected with 50% tissue culture inoculum at 37 ℃, Morphological changes of smooth muscle cells were observed under microscope and electron microscope, and the amount of 3H-hydroxyproline synthesized by 3H-proline incorporated into smooth muscle cells was used as an indicator of collagen. The results showed that smooth muscle cells gradually became round, swollen and local necrosis, and other morphological changes were observed 24h after infection; collagen content of smooth muscle cells in 24, 36 and 48 hours after infection significantly decreased compared with the control group. It is suggested that human herpes simplex virus type 2 may act as a local damage factor, inhibit the synthesis of collagen in vascular smooth muscle cells, impair the smooth muscle cells and participate in the pathogenesis of atherosclerosis.
