目的:探讨Survivin、Bax基因在白毛藤水提液诱导人卵巢癌SK-OV3细胞凋亡中的作用。方法:不同浓度白毛藤水提液作用于SKOV3细胞,荧光显微镜观察SKOV3细胞凋亡作用,RT-PCR法检测Survivin、Bax基因表达量的变化。结果:白毛藤水提液能诱导人卵巢癌细胞凋亡,16mg/mL时凋亡率为(26.06±9.89)%,与对照组(2.24±1.41)%比较差异有统计学意义,P<0.05;白毛藤水提液能上调Bax和降低Survivin表达,实验组与对照组比较差异有统计学意义,P<0.01。结论:白毛藤水提液能促进SKOV3细胞凋亡,其机制可能与激活Bax和抑制Survivin的基因表达有关。
OBJECTIVE: To investigate the role of Survivin and Bax in the apoptosis of human ovarian cancer SK-OV3 cells induced by aqueous extract of Cortex Fragrans. METHODS: SKOV3 cells were treated with different concentrations of aqueous extract from Poncirus above. The apoptosis of SKOV3 cells was observed by fluorescence microscopy. The expression of Survivin and Bax gene was detected by RT-PCR. Results: Aqueous extract of Cortex Fragrans could induce the apoptosis of human ovarian cancer cells. The apoptotic rate was (26.06 ± 9.89)% at 16mg / mL, which was significantly different from that of control group (2.24 ± 1.41)%, P < 0.05; The aqueous extract of white vine can upregulate Bax and reduce the expression of Survivin, the experimental group and the control group, the difference was statistically significant, P <0.01. Conclusion: Aqueous extract of white vine can promote apoptosis of SKOV3 cells, the mechanism may be related to activation of Bax and inhibition of Survivin gene expression.
