BATF、RORγt及IL-17在哮喘小鼠脾组织中的表达及意义

目的探讨B细胞活化转录因子(BATF)、维甲酸相关核孤儿受体γt(RORγt)、白细胞介素-17(IL-17)在哮喘小鼠脾组织中的表达及意义。方法将15只小鼠随机分为3组:生理盐水(NS)组、哮喘(AS)组和地塞米松(DXM)组,每组5只。利用卵清蛋白(OVA)造模。收集支气管肺泡灌洗液(BALF)行白细胞分类计数;HE染色观察肺组织病理改变;酶联免疫吸附测定法(ELISA)检测脾组织匀浆上清液IL-17的质量浓度;实时荧光定量聚合酶链反应(RT-PCR)检测脾组织中BATF、RORγt mRNA的表达量;并对上述指标的相关性进行分析。结果 1)与AS组相比,DXM组小鼠BALF白细胞(WBC)总数、中性粒细胞(NEU)数和嗜酸性粒细胞(EOS)数明显降低(P<0.01)。2)DXM组肺部炎症较AS组明显减轻。3)与AS组相比,DXM组脾组织匀浆上清液IL-17的质量浓度减低(P<0.05)。4)与AS组相比,DXM组脾组织中BATF、RORγt mRNA表达量减少(P<0.01)。5)脾组织匀浆上清液IL-17的质量浓度与BALF中WBC总数、NEU数、EOS数呈正相关(r=0.838、0.737、0.882,P均<0.005);脾组织BATF mRNA的表达量与RORγt mRNA表达量、脾组织匀浆上清液IL-17的质量浓度呈正相关(r=0.807、0.833,P均<0.005);脾组织RORγt mRNA的表达量与脾组织匀浆上清液IL-17的质量浓度呈正相关(r=0.890,P<0.005)。结论 1)在哮喘小鼠脾组织中BATF与RORγt可能共同调节IL-17的分泌,并通过IL-17的致炎作用参与哮喘气道炎症的发生发展。2)DXM可能通过BATF、RORγt途径部分阻止脾源性Th17细胞的分化,减少IL-17的分泌。 Objective To investigate the expression and significance of BATF, RORγt and IL-17 in asthmatic mice. Methods Fifteen mice were randomly divided into three groups: NS group, AS group and DXM group, with 5 mice in each group. Modeling with ovalbumin (OVA). The bronchoalveolar lavage fluid (BALF) was collected for leukocyte sorting and counting; HE staining was used to observe the pathological changes of the lung tissue; the concentration of IL-17 in the supernatant of the spleen tissue was detected by enzyme-linked immunosorbent assay (ELISA); the real- The expression of BATF and RORγt mRNA in spleen tissue was detected by enzyme linked immunosorbent assay (RT-PCR). The correlation between the above parameters was also analyzed. Results 1) Compared with AS group, the number of BALF leucocyte (WBC), neutrophil (NEU) and eosinophil (EOS) in DXM group were significantly decreased (P <0.01). 2) The lung inflammation in DXM group was significantly lower than that in AS group. 3) Compared with AS group, the concentration of IL-17 in the supernatant of the spleen tissue of DXM group decreased (P <0.05). 4) Compared with AS group, the expression of BATF and RORγt mRNA in the spleen of DXM group decreased (P <0.01). 5) The concentration of IL-17 in supernatant of spleen tissue was positively correlated with the total number of WBC, NEU and EOS in BALF (r = 0.838,0.737,0.882, P <0.005). The expression of BATF mRNA in spleen tissue The expression of RORγt mRNA was positively correlated with the concentration of IL-17 in supernatant of spleen (r = 0.807,0.833, P <0.005). The expression of RORγt mRNA in spleen tissue was positively correlated with the level of IL-17 -17 mass concentration was positively correlated (r = 0.890, P <0.005). Conclusions1) BATF and RORγt may regulate the secretion of IL-17 in the spleen of asthmatic mice and participate in the development of airway inflammation in asthma through IL-17. 2) DXM may partially inhibit the differentiation of spleen-derived Th17 cells and decrease the secretion of IL-17 through the BATF and RORγt pathways.
