脑源性神经营养因子Val66Met与红水河流域壮族长寿家系血糖水平的相关性研究

目的探讨红水河流域壮族长寿家系(HLF)脑源性神经营养因子(BDNF)Val66Met(rs6265)多态性与血糖水平的相关性。方法采用ARMS-PCR对HLF(n=1531)rs6265位点进行基因分型,并与当地非长寿家系(HNLF,n=600)及外地区(平果)长寿家系(PLF,n=662)和非长寿家系(PNLF,n=545)进行比较,分析基因型与血糖水平的关系及其在长寿中的作用。结果 (1)HLF rs6265等位基因G和A频率分别为56.3%和43.7%,与其他家系的频率分布差异有统计学意义(P=0.024)。总体上,平果地区A等位基因频率高于红水河流域,PLF中A的频率有低于PNLF的趋势,而HLF A频率则有高于HNLF的趋势;HLF基因型频率分别为GG 31.2%、GA 50.2%、AA 18.6%,与其他对照家系比较差异无统计学意义。(2)PLF GA/AA基因型携带者的血糖水平低于GG携带者(P=0.038)。(3)多元回归分析未发现各家系中血糖水平与基因型的相关性。结论 BDNF Val66Met多态性在红水河地区的分布有异于外地区,可能是该流域长寿的遗传学基础之一,但与血糖水平的关系不大。 Objective To investigate the relationship between Val66Met (rs6265) polymorphism of brain derived neurotrophic factor (BDNF) and blood glucose in Zhuang longevity family (HLF) in Hongshuihe river basin. Methods The rs6265 locus of HLF (n = 1531) was genotyped by ARMS-PCR and was compared with the local non-longevity pedigrees (HNLF, n = 600) and the extended longevity pedigrees (PLF, n = 662) Non-longevous families (PNLF, n = 545) were compared to analyze the relationship between genotypes and blood glucose levels and their role in longevity. Results (1) The frequencies of G and A alleles of HLF rs6265 were 56.3% and 43.7%, respectively, which were significantly different from those of other families (P = 0.024). In general, the frequency of allele A in Pingguo region was higher than that in Hongshui River basin. The frequency of A in PLF was lower than that of PNLF, while the frequency of HLF A was higher than that of HNLF. The frequencies of HLF genotypes were GG 31.2 %, GA 50.2%, AA 18.6%, no significant difference compared with other control pedigrees. (2) The blood glucose level of PLF GA / AA genotype carriers was lower than that of GG carriers (P = 0.038). (3) Multiple regression analysis found no correlation between the blood glucose level and genotype in each lineage. Conclusion The distribution of BDNF Val66Met polymorphism in the Red River region is different from that in the outer region, which may be one of the genetic basis of longevity of the basin, but not related to the blood glucose level.
