目的建立高效液相色谱法快速测定通宣理肺浓缩丸中柚皮苷、橙皮苷、新橙皮苷、迷迭香酸、黄芩苷及甘草酸的含量。方法采用Agilent Porlshell 120 SB-C18(4.6 mm×250 mm,2.7μm)色谱柱,以体积分数70%乙腈为流动相A-0.2%磷酸水溶液B为流动相进行梯度洗脱(0~20 min,25%A→30%A;20~25 min,30%A→95%A;30~31 min,95%A→25%A;31~40min,25%A),流速1.0 mL·min-1,柱温为30℃,柚皮苷、橙皮苷、新橙皮苷检测波长为283 nm(0~10.8 min)、迷迭香酸检测波长为330 nm(10.9~12 min)、黄芩苷检测波长为276 nm(12.1~20 min)、甘草酸检测波长为250 nm(20.1~45 min)。结果柚皮苷、橙皮苷、新橙皮苷、迷迭香酸、黄芩苷及甘草酸的线性范围分别为0.005 69~0.853 2μg、0.003 25~0.487 9μg、0.003 65~0.547 2μg、0.000 51~0.076 2μg、0.007 96~1.193 9μg、0.005 58~0.837 7μg(r≥0.999 0);平均回收率(n=6)分别为98.02%(RSD=1.7%),96.53%(RSD=1.4%),96.44%(RSD=1.3%),96.74%(RSD=1.7%),97.56%(RSD=1.2%),98.57%(RSD=2.0%)。结论所建立的高效液相色谱法可同时测定通宣理肺丸中柚皮苷、橙皮苷、新橙皮苷、迷迭香酸、黄芩苷及甘草酸的含量。
OBJECTIVE To establish a rapid HPLC method for the determination of naringin, hesperidin, neohesperidin, rosmarinic acid, baicalin and glycyrrhizic acid in Tongxuanfeifei Pill. Methods A gradient elution was carried out on a Agilent Porlshell 120 SB-C18 column (4.6 mm × 250 mm, 2.7 μm) with a mobile phase of 70% acetonitrile as mobile phase A-0.2% phosphoric acid solution (0-20 min, 25% A → 30% A; 20-25 min, 30% A → 95% A; 30-31 min, 95% A → 25% A; 31-40 min, 25% , The column temperature was 30 ℃, the detection wavelength of naringin, hesperidin, neohesperidin was 283 nm (0 ~ 10.8 min), the detection wavelength of rosmarinic acid was 330 nm (10.9 ~ 12 min) Wavelength of 276 nm (12.1 ~ 20 min), glycyrrhizic acid detection wavelength of 250 nm (20.1 ~ 45 min). Results The linear range of naringin, hesperidin, neohesperidin, rosmarinic acid, baicalin and glycyrrhizin were 0.005 69 ~ 0.853 2μg, 0.003 25 ~ 0.487 9μg, 0.003 65 ~ 0.547 2μg, 0.000 51 ~ The mean recoveries (n = 6) were 98.02% (RSD = 1.7%), 96.53% (RSD = 1.4%) and 96.44 % (RSD = 1.3%), 96.74% (RSD = 1.7%), 97.56% (RSD = 1.2%), 98.57% (RSD = 2.0%). Conclusion The established HPLC method can simultaneously determine the contents of naringin, hesperidin, neohesperidin, rosmarinic acid, baicalin and glycyrrhizic acid in Tongxuanlifei pill.
