以“粤甜13号”单倍体种子为材料,利用不同浓度秋水仙素直接浸泡单倍体种子,同时设置加倍时间及加倍温度两个因素,并对单倍体浸种方法进行优化。结果表明,秋水仙素浓度、加倍时间和加倍温度均会影响甜玉米单倍体加倍效果,通过秋水仙素直接浸泡甜玉米单倍体种子,可显著提高甜玉米单倍体加倍率。如不考虑各因素间互作,最适加倍浓度、处理时间及加倍温度分别为0.02%、30 h、32℃;若考虑各因素间互作,0.02%浓度,24℃处理32 h可获得较高的单倍体结实率,药害率低,且均优于自然加倍和常规秋水仙素处理。
The haploid seeds of “Yue Tian No. 13” were used as materials, the haploid seeds were directly soaked with colchicine at different concentrations, and the doubling time and doubling temperature were set at the same time. The haploid soaking method was optimized. The results showed that colchicine concentration, doubling time and doubling temperature both affected the doubling effect of sweet maize haploid. Directly soaking colchicine seeds with colchicine could significantly increase haploid doubling rate of sweet corn. If we do not consider the interactions among factors, the optimum concentration, treatment time and doubling temperature are 0.02%, 30 h and 32 ℃, respectively. If we consider the interaction between various factors, 0.02% concentration and 24 ℃ treatment for 32 h High haploid solid rate, low rate of injury, and are better than natural doubling and conventional colchicine treatment.
