作者制备了12株抗仙台病毒单克隆抗体,并对其特异性、生物学功能和同种型等进行了研究。以100μg纯化的仙台病毒作为抗原加弗氏完全佐剂,腹腔注射Lou鼠,14和30天后分别以相同剂量的抗原加弗氏不完全佐剂再次注射,约10天后以放射免疫法(RIA)测试抗仙台病毒抗体滴度。取抗体滴度于1:200以上的Lou鼠,以200μg抗原加强免疫一次,4天后取其脾细胞与FO骨髓瘤细胞或Y3骨髓瘤细胞融合。经RIA筛选出
The authors prepared 12 anti-Sendai virus monoclonal antibodies, and its specificity, biological functions and isoforms were studied. Lou mice were injected intraperitoneally with 100 μg of purified Sendai virus as antigen plus Freund’s complete adjuvant. After 14 and 30 days, they were re-injected with the same dose of antigen plus incomplete Freund’s adjuvant. After about 10 days, radioimmunoassay (RIA) Test anti-Sendai virus antibody titers. Take Lou mice with antibody titer of 1: 200 or more, and immunize with 200 μg of antigen once. After 4 days, the spleen cells are fused with FO myeloma cells or Y3 myeloma cells. Filter out by RIA
