目的研究转导CⅡTA基因肿瘤细胞的生物学特性改变。方法以RT-PCR和流式细胞术分别检测克隆转导CⅡTA基因的肿瘤细胞中CⅡTA基因的表达。在光镜和电镜下观察细胞形态和结构变化。结并转导CⅡTA基因后,肿瘤细胞中CⅡTA基因得以有效表达,肿瘤细胞形态和细胞内的超微结构发生改变,细胞生长受限,IFN-7可诱导转导CⅡTA基因的肿瘤细胞死亡。结论转导CⅡTA基因能改变肿瘤细胞的生物学特性,抑制肿瘤细胞生长。
Objective To study the biological characteristics of tumor cells transduced with CIITA gene. Methods The expression of CIITA gene in tumor cells transfected with CIITA gene was detected by RT-PCR and flow cytometry, respectively. The morphology and structural changes of the cells were observed under light microscope and electron microscope. After CIITA gene was transduced and transduced, the CIITA gene in tumor cells could be effectively expressed. The morphology and intracellular ultrastructure of tumor cells were changed. The growth of cells was limited, and IFN-7 could induce the death of tumor cells transduced with CIITA gene. Conclusion Transduction of CIITA gene can alter the biological characteristics of tumor cells and inhibit the growth of tumor cells.
