目的 探索miR-199a,HIF-1α在银屑病中的表达及其对表皮细胞增殖的影响.方法 首先检测了27例寻常性银屑病患者皮损区和19例健康志愿者皮肤的miR-199a,HIF-1α的表达差异,分析二者之间的相关性;然后在HaCaT细胞中转染miR-199a mimics,用Western blot检测HIF-1 α的蛋白表达水平;用双荧光素酶报告基因实验验证miR-199a与HIF-1α的靶向结合关系;最后用MTT法检测miR-199a,HIF-1α对HaCaT细胞增殖的影响.结果 银屑病组织中miR-199a的表达下调,HIF-1 α的表达上调,且两者存在负相关性;在HaCaT细胞中转染miR-199a mimics可下调HIF-1α的表达;HIF-1α可回复miR-199a对HaCaT细胞增殖的抑制作用.结论 miR-199a靶向HIF-1α调控表皮细胞的增殖从而参与银屑病的发病机制.“,”Objective To explore the expression of miR-199a and HIF-1α in psoriatic patients and its effect on proliferation in HaCat keratinocytes.Methods Firstly,27 patients with psoriasis and 19 healthy volunteers were recruited,the expression of miR-199a and HIF-1α were detected using Real-time PCR,and the correlation between them was analyzed.Then we transfected miR-199a mimics into HaCaT cells,the HIF-1α protein expression was detected by Western blot,double luciferase reporter assay was used to verify miR-199a could directly target the 3'UTR region of HIF-1α.Lastly,we use MTT assays to detect the effects of miR-199a and and HIF-1 α on the proliferation of HaCaT cells.Results The expression of miR-199a was down-regulated and the expression of HIF-1 α was up-regulated in psoriasis tissues,and there was a negative correlation between them.miR-199a mimics down-regulated the protein expression of HIF-1α;HIF-1α overexpression restored the inhibitory effect of miR-199a on the proliferation of HaCaT cells.Conclusion MiR-199a targets HIF-1 α to regulate the proliferation of HaCaT cells and participate in the pathogenesis of psoriasis.
