目的观察自然衰老大鼠海马组织Nestin和Ki-67蛋白表达变化及补肾方作用,探索衰老与海马NSCs变化的相关性及补肾方延缓衰老的作用机理。方法 SD雄性大鼠,SPF级,分为老年组、左归丸组、右归丸组和青年组,共4组。常规喂养饲料,自由饮水。老年组大鼠常规喂养至24月龄;左归丸组和右归丸组大鼠自20月龄始,每天分别喂以含有左归丸和右归丸生药7.5g/kg的饲料,连续4个月。采用Western blot法检测大鼠海马组织Nestin和Ki-67蛋白表达。结果与青年组大鼠比较,老年组大鼠海马组织Nestin和Ki-67蛋白表达均下调(P<0.05);与老年组比较,左归丸组和右归丸组大鼠海马组织Nestin和Ki-67蛋白表达均上调(P<0.05)。结论自然衰老大鼠海马NSCs分裂和增殖能力减弱,左归丸和右归丸通过上调海马组织Nestin和Ki-67蛋白的表达,进而促进NSCs的增殖可能是其延缓衰老的机理之一。
Objective To observe the changes of Nestin and Ki-67 protein in hippocampus of rats with natural aging and the effect of Bushen Recipe on exploring the correlation between aging and the changes of NSCs in hippocampus and the mechanism of Bushen Fang delaying aging. Methods Male Sprague-Dawley rats were divided into 4 groups: old group, Zuoguiwan group, Yougui pill group and youth group. Routine feeding, free to drink water. The rats in the aged group were fed to 24 months old routinely. The rats in Zuogui Pill group and Yougui Pill group were fed with 7.5g / kg feed of Zuogui Pill and Yougui Pill respectively at the beginning of 20 months, month. Western blot was used to detect the expression of Nestin and Ki-67 in hippocampus of rats. Results Compared with the young group, the expressions of Nestin and Ki-67 in the hippocampus of the aged group were significantly decreased (P <0.05). Compared with the old group, the expressions of Nestin and Ki in hippocampus of Zuoguiwan and Yougui Wan -67 protein expression were increased (P <0.05). Conclusions The ability of NSCs to differentiate and proliferate in hippocampus of spontaneously aged rats is weakened. Zuogui pill and Yougui pill may be one of the mechanisms of anti-aging by up-regulating the expression of Nestin and Ki-67 in hippocampus and further promoting the proliferation of NSCs.
