目的构建野生小鼠抗肺肿瘤基因差异表达文库。方法应用抑制性消减杂交(SSH)技术对TAF1小鼠与A/wy近交系小鼠中差异表达的抗肺肿瘤基因进行筛选,构建消减cDNA文库,并对部分克隆进行核苷酸序列分析,利用BLAST检索分析其可能代表的基因。结果166个差异表达克隆测序后进行BLAST分析,其中134个克隆与87个已知小鼠的mRNA或cDNA序列同源,32个克隆与14个DNA序列同源;87个已知基因按功能分为7大类。其他32个克隆中,20个克隆与9个鼠DNA序列同源,其中可能含有新基因,2个克隆与1个细菌序列同源,3个克隆为克隆载体。结论应用SSH技术成功构建了野生小鼠抗肺肿瘤基因差异表达文库。
Objective To construct a differential expression library of lung cancer gene in wild mice. Methods The anti-lung tumor genes differentially expressed in TAF1 mice and A / wy inbred mice were screened by SSH and the subtracted cDNA library was constructed. Nucleotide sequences of some clones were analyzed. BLAST search was used to analyze the possible genes. Results A total of 166 differential expression clones were sequenced and analyzed by BLAST. 134 clones were homologous to the mRNA or cDNA sequences of 87 known mice, 32 clones were homologous to 14 DNA sequences, and 87 known genes were classified by function For seven categories. Among the other 32 clones, 20 clones were homologous to 9 murine DNA sequences, which may contain new genes, 2 clones were homologous to 1 bacterial sequence, and 3 clones were cloning vectors. Conclusion The wild type mouse lung cancer gene differential expression library was constructed successfully by SSH technology.
