目的:建立大鼠甲基苯丙胺(methaphetamine,MA)慢性中毒模型,探讨MA长期应用对脑纹状体区的损伤作用。方法:Wistar大鼠30只,雄性,体重180~220 g,随机分为对照组(A)和实验组(B)。B组从小剂量开始腹腔注射MA,逐渐加量,共用4周;A组腹腔注射等量生理氯化钠溶液。电子体温计监测体温变化;TUNEL法观察纹状体内神经元凋亡情况;高效液相色谱检测纹状体内多巴胺(dopamine,DA)含量。结果:MA引起大鼠行为活动明显增多;纹状体区神经细胞凋亡数量显著增多及DA含量明显下降。结论:MA慢性中毒对大鼠脑组织纹状体有明显的毒性损伤,可引起大鼠行为改变,纹状体神经元大量凋亡及DA含量显著下降。
OBJECTIVE: To establish chronic poisoning model of methamphetamine (MA) in rats and explore the damaging effect of long-term MA on brain striatum. Methods: Thirty male Wistar rats weighing 180-220 g were randomly divided into control group (A) and experimental group (B). In group B, MA was injected intraperitoneally from a small dose, and gradually added for 4 weeks. In group A, an equal volume of physiological sodium chloride solution was injected intraperitoneally. The changes of body temperature were monitored by electronic thermometer. The apoptosis of neurons in striatum was observed by TUNEL method. The content of dopamine (DA) in striatum was detected by high performance liquid chromatography. Results: MA induced a significant increase in behavioral activity in rats. The number of apoptotic neurons and the content of DA in striatum were significantly decreased. CONCLUSION: Chronic poisoning of MA significantly damages the striatum of rat brain and causes behavioral changes in rat. The apoptosis of striatum neurons and the DA content decrease significantly.
