川芎嗪对大鼠胶原性关节炎及免疫功能的影响

目的:探讨川芎嗪对胶原性关节炎大鼠的影响。方法:采用Ⅱ型胶原诱导胶原性关节炎大鼠模型。随机分为正常组、模型组与川芎嗪(50 mg/kg、100mg/kg)处理组。实验过程中观察胶原性大鼠的一般情况、足爪肿胀度和关节指数的变化。给药结束后第2天,3%巴比妥那麻醉处死大鼠,取足爪组织制备切片观察病理变化;无菌取脾制备脾细胞悬液,用MTT法检测脾T细胞和B细胞增殖反应;制备10%脾组织匀浆用ELSIA法检测脾组织IL-1β、TNF-α和IL-12含量;心脏取血,制备血清用ELSIA法测定血清中抗CⅡ抗体水平。结果:与正常组比较,模型组大鼠关节红肿,表面皮肤发亮充血,关节指数上升,脾T细胞和B细胞增殖反应、血清中抗CⅡ抗体水平和脾组织中IL-1β、TNF-α和IL-12水平明显升高,大鼠足爪皮下组织细胞排列紊乱且有大量炎性细胞浸润及血管增生。与模型组比较,川芎嗪100mg/kg可改善胶原性关节炎大鼠一般状况,明显抑制大鼠足爪肿胀;降低关节指数;抑制脾T细胞和B细胞增殖反应,在570nm的吸光度A分别为0.579±0.017 vs 0.624±0.015,0.263±0.021 vs 0.312±0.017;血清中抗CⅡ抗体水平显著降低,由模型对照组的(0.756±0.020)μg/L降低到(0.579±0.023)μg/L;脾组织中IL-1β〔(80.32±12.25)vs(160.06±20.61)ng/L〕,TNF-α〔(81.26±13.57)vs(123.51±20.34)ng/L〕和IL-12〔(181.05±19.57)vs(260.08±17.52)ng/L〕水平均明显降低;能明显改善大鼠足爪组织病理改变,炎性细胞浸润及血管明显减轻,川芎嗪50 mg/kg对上述指标无明显影响。结论:川芎嗪100 mg/kg可抑制CIA大鼠脾T和B淋巴细胞的增殖反应,降低脾组织中IL-1β、TNF-α和IL-12和血清中抗CⅡ抗体水平,改善足爪组织病理变化,对Ⅱ型胶原诱导的关节炎具有治疗作用。 Objective: To investigate the effect of ligustrazine on collagen-induced arthritis in rats. Methods: collagen type Ⅱ collagen induced arthritis rat model. Randomly divided into normal group, model group and ligustrazine (50 mg / kg, 100mg / kg) treatment group. During the experiment, we observed the general situation of collagen-induced rats, changes of paw swelling and joint index. On the second day after the administration, rats were killed by 3% barbital anesthesia. Tissue sections were obtained from the paw to observe the pathological changes. Spleen cell suspension was prepared by sterile spleen extraction. The proliferation of splenic T cells and B cells were detected by MTT assay The levels of IL-1β, TNF-α and IL-12 in spleen tissue were measured by ELSIA. Blood samples were obtained from the heart and the levels of anti-CⅡ antibody in serum were determined by ELSIA. Results: Compared with the normal group, the model group had swollen joints, shiny and hyperemia on the surface of the skin, increased articular index, proliferation of spleen T cells and B cells, anti-CⅡ antibody levels in serum and IL-1β and TNF-α And IL-12 levels were significantly increased, foot paw tissue cells arranged disorderly and a large number of inflammatory cell infiltration and angiogenesis. Compared with the model group, Ligustrazine 100 mg / kg can improve the general condition of collagen induced arthritis rats, significantly inhibit the swelling of paw in rats, reduce the index of joint, inhibit the proliferation of spleen T cells and B cells, the absorbance A at 570 nm is 0.579 ± 0.017 vs 0.624 ± 0.015, 0.263 ± 0.021 vs 0.312 ± 0.017, respectively. The level of anti-CⅡ antibody in serum was significantly decreased from (0.756 ± 0.020) μg / L to (0.579 ± 0.023) μg / L in model group IL-1β 〔(80.32 ± 12.25) vs (160.06 ± 20.61〕 ng / L〕, TNF-α 〔(81.26 ± 13.57) vs (123.51 ± 20.34〕 ng / L〕 and IL-12 〔(181.05 ± 19.57 ) (260.08 ± 17.52) ng / L〕 significantly reduced the pathological changes of paw tissue, inflammatory cell infiltration and vascular significantly reduced, ligustrazine 50 mg / kg had no significant effect on the above parameters. CONCLUSION: Tetramethylpyrazine 100 mg / kg can inhibit the proliferation of spleen T and B lymphocytes in CIA rats, decrease the levels of IL-1β, TNF-α, IL-12 and anti-CⅡ in serum, Pathological changes, the type II collagen-induced arthritis has a therapeutic effect.
